  • Check difference factor for reversals in literature ( Andre Brown, Early tracking papers)
  • Aim for n=10 in manual counting of pumping
  • Plot reversals over time by setting threshhold and difference → look at amount of positive transitions, average N reversals over total time
  • Plot reversals in different colors ( mutlicolor plot) to be able to tell if analysis is similar to visual observation

  • Manual counting was done by loading in 764 frames of image sequence (due to laptop memory) and counting pharynngeal pumping via counting app and then scaling the pumps to framerate equal to one minute (1800 frames)

  • Figures: everything 12 points or bigger (14), 3,5 inches in width*2, upload figures to lab book (n, t-test)
  • Update log on conversion factor for 2*2 binning with picture of scale in FIJI, take picture of 1*1 Binning with scale and calculate conversion
  • Reversal analysis: change diff of x and y to better fit scale ( 15-30 difference)
  • Count pumping manually in 1*1 Binning image sequences on 2% and 10% Agar
    • Count several worms for a timespan of one minute
